Gene Summary This gene encodes a class III member of the beta tubulin protein family. Beta tubulins are one of two core protein families (alpha and beta tubulins) that heterodimerize and assemble to form microtubules. This protein is primarily expressed in neurons and may be involved in neurogenesis and axon guidance and maintenance. Mutations in this gene are the cause of congenital fibrosis of the extraocular muscles type 3. Alternate splicing results in multiple transcript variants. A pseudogene of this gene is found on chromosome 6. [provided by RefSeq, Oct 2010]

Application

ICC/IF

Immunocytochemistry analysis of HepG2 cells labeling Beta III Tubulin with Mouse anti-Beta III Tubulin antibody at 1/50 dilution. Cells were fixed in 4% paraformaldehyde for 30 minutes, permeabilized with 0.05% Triton X-100 in PBS for 20 minutes, and then blocked with 2% negative goat serum for 30 minutes at room temperature. Cells were then incubated with Mouse anti-Beta III Tubulin antibody (488) at 1/50 dilution in 2% negative goat serum overnight at 4 ℃. Nuclear DNA was labelled in blue with DAPI. beta Tubulin (red) was stained at 1/100 dilution overnight at +4℃. Goat Anti-Rabbit IgG H&L (647) were used as the secondary antibody at 1/1,000 dilution.

IF-P

Immunofluorescence analysis of paraffin-embedded rat brain tissue labeling Beta III Tubulin with Mouse anti-Beta III Tubulin antibody at 1/200 dilution. The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 20 minutes. The tissues were blocked in 10% negative goat serum for 1 hour at room temperature, washed with PBS, and then probed with the primary antibody (green) at 1/200 dilution overnight at 4 ℃, washed with PBS. Nuclei were counterstained with DAPI (blue).

Flow cytometric analysis of SH-SY5Y cells labeling Beta III Tubulin. Cells were fixed and permeabilized. Then incubated for 1 hour at +4℃ with Beta III Tubulin (red, 1ug/ml). Unlabelled sample was used as a control (cells without incubation with primary antibody; black).
