Western blot analysis of Phospho-MDM2 (S166) on different lysates with Rabbit anti-Phospho-MDM2 (S166) antibody at 1/1,000 dilution. Lane 1: A549 cell lysate, Lane 2: A549 treated with λpp for 1 hour cell lysate, Lysates/proteins at 20 µg/Lane. Exposure time: 57 seconds; 4-20% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody at 1/1,000 dilution was used in 5% NFDM/TBST at 4℃ overnight. The encoded protein can promote tumor formation by targeting tumor suppressor proteins, such as p53, for proteasomal degradation. This gene is itself transcriptionally-regulated by p53. Overexpression or amplification of this locus is detected in a variety of different cancers. There is a pseudogene for this gene on chromosome 2.
