Product Profile
Product Name Her2 / ErbB2 antibody (ready-to-use)
Antibody Type Primary Antibodies
Product description Rabbit monoclonal antibody to ErbB2This gene encodes a member of the epidermal growth factor (EGF) receptor family of receptor tyrosine kinases. This protein has no ligand binding domain of its own and therefore cannot bind growth factors. However, it does bind tightly to other ligand-bound EGF receptor family members to form a heterodimer, stabilizing ligand binding and enhancing kinase-mediated activation of downstream signalling pathways, such as those involving mitogen-activated protein kinase and phosphatidylinositol-3 kinase. Allelic variations at amino acid positions 654 and 655 of isoform a (positions 624 and 625 of isoform b) have been reported, with the most common allele, Ile654/Ile655, shown here. Amplification and/or overexpression of this gene has been reported in numerous cancers, including breast and ovarian tumors. Alternative splicing results in several additional transcript variants, some encoding different isoforms and others that have not been fully characterized. [provided by RefSeq]
Immunogen Recombinant protein encoding extracellular domain of human c-erbB-2.
Key Feature
Clonality Monoclonal
Isotype IgG
Clone Number SP3
Host Species Rabbit
Tested Applications IHC-P
We suggest an incubation period of 30 minutes at room temperature. However, depending upon the fixation conditions and the staining system employed, optimal incubation should be determined by the user. Formalin fixed paraffin embedded tissue sections require high temperature antigen unmasking with 10 mM citrate buffer, pH 6.0 prior to immunostaining.:

Tissue Specificity This antibody recognizes a c-erbB-2 protein, which is a receptor tyrosine kinase of the c-erbB family. It is closely related in structure to the epidermal growth factor receptor. c-erbB-2 oncoprotein is detectable in a proportion of breast and other adenocarcinomas, as well as transitional cell carcinomas. In the case of breast cancer, expression determined by immunohistochemistry has been shown to be associated with poor prognosis. This antibody crossreacts with Human.
Cellular Localization Cell membrane
