Product Profile
Product Name Huntingtin (neoepitope 586) antibody
Antibody Type Primary Antibodies
Product description Rabbit polyclonal antibody to Huntingtin (neoepitope 586)Huntingtin is a disease gene linked to Huntington's disease, a neurodegenerative disorder characterized by loss of striatal neurons. This is thought to be caused by an expanded, unstable trinucleotide repeat in the huntingtin gene, which translates as a polyglutamine repeat in the protein product. A fairly broad range in the number of trinucleotide repeats has been identified in normal controls, and repeat numbers in excess of 40 have been described as pathological. The huntingtin locus is large, spanning 180 kb and consisting of 67 exons. The huntingtin gene is widely expressed and is required for normal development. It is expressed as 2 alternatively polyadenylated forms displaying different relative abundance in various fetal and adult tissues. The larger transcript is approximately 13.7 kb and is expressed predominantly in adult and fetal brain whereas the smaller transcript of approximately 10.3 kb is more widely expressed. The genetic defect leading to Huntington's disease may no

Application

Application

ICC/IF analysis of Caspase cleaved Htt (green) in 293T cells transfected with Htt23Q and Htt148Q stop constructs ending in amino acid 586 using Huntingtin (neoepitope 586) antibody at a dilution of 1:50 (green). Nuclei (blue) were stained with Hoechst dye.

Application

IHC analysis of tissues from either transgenic HD mice of the YAC128 line (left) or their WT littermates (right). Striatum (top panels) and cortex (bottom panels) tissues were stained using Huntingtin (neoepitope 586) antibody at a dilution of 1:50 (green).

Application

Sandwich ELISA was performed with a Huntingtin monoclonal antibody and Huntingtin (neoepitope 586) antibody to determine the antigen concentration of the Huntingtin cleavage products. The curve represents a dose response for neo586 in 293T cells overexpressing the Htt construct.

Application

WB analysis of endogenous Htt lysates with or without different caspase activity (lanes 1-5) and overexpressed recombinant Htt fragment lysates (lanes 6-9) (20 ug per well) using pan-Huntingtin antibody (top, N-terminal HTT) or Huntingtin (neoepitope 586) antibody at a dilution of 1:500 (bottom, neoHtt586).
