ELISA: Use at an assay dependent dilution. IHC-Fr: Use at an assay dependent dilution. IP: Use at an assay dependent dilution. WB: Use at a dilution of 1/1000, this dilution is determined using a concentrated cell culture media from a stimulated human cell line, an alkaline phosphatase conjugated secondary antibody and BCIP/NBT as the substrate. Higher antibody concentrations may be necessary for non-human samples. By blotting against the reduced protein, the antibody reacts with bands at 58 kDa (proenzyme), 45 kDa (active), and a series of further cleaved active forms. Since MMP23 levels in quiescent cells and tissues are minimal, mitogen stimulation or protein concentration is often needed to visualize the bands by immunoblotting. Treatment of the cells with the phorbal ester TPA or IL2 stimulates the production of MMP23 in some cell types. In addition, cell types differ greatly in the quantity of MMP23 produced. Even though MMP23 is localized within the cell, it may also be seen in conditioned media from cells in culture. Optimal dilutions/concentrations should be determined by the end user.:
