Gene Summary The pyruvate dehydrogenase (PDH) complex is a nuclear-encoded mitochondrial multienzyme complex that catalyzes the overall conversion of pyruvate to acetyl-CoA and CO(2), and provides the primary link between glycolysis and the tricarboxylic acid (TCA) cycle. The PDH complex is composed of multiple copies of three enzymatic components: pyruvate dehydrogenase (E1), dihydrolipoamide acetyltransferase (E2) and lipoamide dehydrogenase (E3). The E1 enzyme is a heterotetramer of two alpha and two beta subunits. This gene encodes the E1 alpha 1 subunit containing the E1 active site, and plays a key role in the function of the PDH complex. Mutations in this gene are associated with pyruvate dehydrogenase E1-alpha deficiency and X-linked Leigh syndrome. Alternatively spliced transcript variants encoding different isoforms have been found for this gene.[provided by RefSeq, Mar 2010]

Western blot analysis of extracts from various samples, using Phospho-PDHA1/2 (Ser293/Ser291) Antibody. Lane 1: Pc12 cells(heat-shock treatment), blocked with antigen-specific peptides. Lane 2: Pc12 cells(heat-shock treatment). Lane 3: 3t3-l1 cells(heat-shock treatment). Lane 4: Hepg2 cells(heat-shock treatment). Lane 5: Hela cells(heat-shock treatment).

IHC

Rabbit polyclonal antibody to Phospho-PDHA1/2 (Ser293/Ser291) at 1/100 staining Human colorectal cancer by IHC-P. The sample was formaldehyde fixed and a heat mediated antigen retrieval step in citrate buffer was performed. The sample was then blocked and incubated with the primary antibody at 4°C overnight. An HRP conjugated anti-Rabbit antibody was used as the secondary antibody.

ICC/IF

Rabbit polyclonal antibody to Phospho-PDHA1/2 (Ser293/Ser291) staining Hela cells(4h of LPS treatment) by IF/ICC. The samples were fixed with PFA and permeabilized in 0.1% Triton X-100,then blocked in 10% serum for 45 minutes at 25°C. Samples were then incubated with primary Ab(1:200) and mouse anti-beta tubulin Ab(1:200) for 1 hour at 37°C. An AlexaFluor594 conjugated goat anti-rabbit IgG(H+L) Ab(Red) and an AlexaFluor488 conjugated goat anti-mouse IgG(H+L) Ab(Green) were used as the secondary antibody. The nuclear counter stain is DAPI(blue).
