Western blot analysis of ARNTL2 using anti-ARNTL2 antibody. The sample well of each lane was loaded with 30 ug of sample under reducing conditions. Lane 1: human PC-3 whole cell lysates, Lane 2: human A549 whole cell lysates, Lane 3: human HepG2 whole cell lysates, Lane 4: rat ovary tissue lysates, Lane 5: mouse lung tissue lysates, Lane 6: mouse testis tissue lysates. After electrophoresis, proteins were transferred to a membrane. Then the membrane was incubated with rabbit anti-ARNTL2 antigen affinity purified polyclonal antibody at a dilution of 1:1000 and probed with a goat anti-rabbit IgG-HRP secondary antibody. The signal is developed using ECL Plus Western Blotting Substrate.IHC
IHC analysis of ARNTL2 using anti-ARNTL2 antibody. ARNTL2 was detected in a paraffin-embedded section of human glioma cancer tissue. Biotinylated goat anti-rabbit IgG was used as secondary antibody. The tissue section was incubated with rabbit anti-ARNTL2 Antibody at a dilution of 1:200 and developed using Strepavidin-Biotin-Complex (SABC) with DAB as the chromogen.IHC

Flow Cytometry analysis of HL-60 cells using anti-ARNTL2 antibody. Overlay histogram showing HL-60 cells stained with anti-ARNTL2 antibody (Blue line). To facilitate intracellular staining, cells were fixed with 4% paraformaldehyde and permeabilized with permeabilization buffer. The cells were blocked with 10% normal goat serum. And then incubated with rabbit anti-ARNTL2 Antibody at 1:100 dilution for 30 min at 20°C. DyLight®488 conjugated goat anti-rabbit IgG was used as secondary antibody at 1:100 dilution for 30 minutes at 20°C. Isotype control antibody (Green line) was rabbit IgG at 1:100 dilution used under the same conditions. | Gene Summary | This gene encodes a basic helix-loop-helix transcription factor belonging to the PAS (PER, ARNT, SIM) superfamily. The PAS proteins play important roles in adaptation to low atmospheric and cellular oxygen levels, exposure to certain environmental pollutants, and diurnal oscillations in light and temperature. This protein forms a transcriptionally active heterodimer with the circadian CLOCK protein, the structurally related MOP4, and hypoxia-inducible factors, such as HIF1alpha. Consistent with its role as a biologically relevant partner of circadian and hypoxia factors, this protein is coexpressed in regions of the brain such as the thalamus, hypothalamus, and amygdala.
