WB
Western blot analysis of various lysates using IL-6 Rabbit mAb at 1:5000 dilution incubated overnight at 4℃. RAW 264.7 cells and NR8383 cells were treated with LPS (1μg/mL) and BFA (300ng/mL) for 8 hours. Secondary antibody: HRP-conjugated Goat anti-Rabbit IgG (H+L) at 1:10000 dilution. Lysates/proteins: 30 μg per lane. Blocking buffer: 3% nonfat dry milk in TBST. Detection: ECL Basic Kit. Exposure time: 45s.
ICC/IF
Confocal imaging of RAW 264.7 cells (treated with LPS and BFA) and RAW 264.7 cells (untreated) using IL-6 Rabbit mAb (dilution 1:200) followed by a further incubation with Cy3-conjugated Goat Anti Rabbit IgG (H+L) (dilution 1:500) (Red). DAPI was used for nuclear staining (Blue). Objective: 100x.
IP
Immunoprecipitation of IL-6 from 1000 µg extracts of NR8383 cells treated with LPS (100 ng/ml, 24 h) and BFA (1 µg/ml, 6 h) was performed using 1 µg of IL-6 Rabbit mAb. Rabbit Control IgG was used to precipitate the Control IgG sample. IP samples were eluted with 1x reducing Laemmli Buffer. The Input lane represents 10% of the total input. Western blot analysis of immunoprecipitates was conducted using IL-6 Rabbit mAb at a dilution of 1:10000.
