PTP-PEST
Gene Full Name protein tyrosine phosphatase non-receptor type 12
Gene Summary The protein encoded by this gene is a member of the protein tyrosine phosphatase (PTP) family. PTPs are signaling molecules that regulate a variety of cellular processes including cell growth, differentiation, mitotic cycle, and oncogenic transformation. This PTP contains a C-terminal PEST motif, which serves as a protein-protein interaction domain, and may regulate protein intracellular half-life. This PTP was found to bind and dephosphorylate the product of the oncogene c-ABL and thus may play a role in oncogenesis. This PTP was also shown to interact with, and dephosphorylate, various products related to cytoskeletal structure and cell adhesion, such as p130 (Cas), CAKbeta/PTK2B, PSTPIP1, and paxillin. This suggests it has a regulatory role in controlling cell shape and mobility. Alternative splicing results in multiple transcript variants encoding distinct isoforms. [provided by RefSeq, Oct 2008]

Application

WB

Western blot analysis of lysates from various samples, using PTPN12 Ab. Lane 1: HepG2 cells(heat-shock treatment), blocked with antigen-specific peptides. Lane 2: HepG2 cells(heat-shock treatment). Lane 3: Hela cells(heat-shock treatment).

IHC

Rabbit polyclonal antibody to PTPN12 at 1/100 staining rat brain tissue by IHC-P. The sample was formaldehyde fixed and a heat mediated antigen retrieval step in citrate buffer was performed. The sample was then blocked and incubated with the primary Ab at 4°C overnight. An HRP conjugated anti-rabbit Ab was used as the secondary Ab.

ICC/IF

Rabbit polyclonal antibody to PTPN12 staining Hela cells by IF/ICC. The samples were fixed with PFA and permeabilized in 0.1% Triton X-100, then blocked in 10% serum for 45 minutes at 25°C. Samples were then incubated with primary Ab and mouse anti-beta tubulin Ab for 1 hour at 37°C. An AlexaFluor594 conjugated goat anti-rabbit IgG Ab(Red) and an AlexaFluor488 conjugated goat anti-mouse IgG Ab(Green) were used as the secondary Ab. The nuclear counter stain is DAPI (blue).
