Gene Full Name poly(ADP-ribose) polymerase 1
Gene Summary This gene encodes a chromatin-associated enzyme, poly(ADP-ribosyl)transferase, which modifies various nuclear proteins by poly(ADP-ribosyl)ation. The modification is dependent on DNA and is involved in the regulation of various important cellular processes such as differentiation, proliferation, and tumor transformation and also in the regulation of the molecular events involved in the recovery of cell from DNA damage. In addition, this enzyme may be the site of mutation in Fanconi anemia, and may participate in the pathophysiology of type I diabetes. [provided by RefSeq, Jul 2008]

Application

WB

Western blot analysis of PARP1 using anti-PARP1 antibody. The sample well of each lane was loaded with 30 ug of sample under reducing conditions. Lane 1: human Hela whole cell lysates, Lane 2: human HepG2 whole cell lysates, Lane 3: human COLO-320 whole cell lysates, Lane 4: human Jurkat whole cell lysates, Lane 5: rat PC-12 whole cell lysates, Lane 6: mouse NIH/3T3 whole cell lysates, Lane 7: mouse HEPA1-6 whole whole cell lysates. After electrophoresis, proteins were transferred to a membrane. Then the membrane was incubated with rabbit anti-PARP1 antigen affinity purified polyclonal antibody at a dilution of 1:1000 and probed with a goat anti-rabbit IgG-HRP secondary antibody. The signal is developed using ECL Plus Western Blotting Substrate. A specific band was detected for PARP1 at approximately 113 kDa. The expected band size for PARP1 is at 113 kDa.

IHC

IHC analysis of PARP1 using anti-PARP1 antibody. PARP1 was detected in a paraffin-embedded section of mouse intestine tissue. Biotinylated goat anti-rabbit IgG was used as secondary antibody. The tissue section was incubated with rabbit anti-PARP1 Antibody at a dilution of 1:200 and developed using Strepavidin-Biotin-Complex (SABC) with DAB as the chromogen.

IHC

IHC analysis of PARP1 using anti-PARP1 antibody . PARP1 was detected in a paraffin-embedded section of human placenta tissue.

ICC/IF

IF analysis using anti- PARP antibody. detected in paraffin-embedded section of human breast cancer tissue. The tissue section were stained using the cy3-conjugated Anti-rabbit IgG Secondary Antibody (red) and counterstained with DAPI (blue).

ICC/IF

ICC/IF analysis of PARP1 using anti-PARP1 antibody . PARP1 was detected in an immunocytochemical section of U2OS cells. The section was incubated with rabbit anti-PARP1 Antibody at a dilution of 1:100. Fluoro488 Conjugated Goat Anti-Rabbit IgG (Green) was used as secondary antibody. The section was counterstained with DAPI (Blue).

FC

Flow Cytometry analysis of A431 cells using anti-PARP1 antibody. Overlay histogram showing A431 cells stained with PB9309 (Blue line). To facilitate intracellular staining, cells were fixed with 4% paraformaldehyde and permeabilized with permeabilization buffer. The cells were blocked with 10% normal goat serum. And then incubated with rabbit anti-PARP1 Antibody at 1:100 dilution for 30 min at 20°C. Fluoro488 conjugated goat anti-rabbit IgG was used as secondary antibody at 1:100 dilution for 30 minutes at 20°C. Isotype control antibody (Green line) was rabbit IgG at 1:100 dilution used under the same conditions. Unlabelled sample without incubation with primary antibody and secondary antibody (Red line) was used as a blank control.
