Gene Synonyms hMLKL
Gene Full Name mixed lineage kinase domain like pseudokinase
Gene Summary This gene belongs to the protein kinase superfamily. The encoded protein contains a protein kinase-like domain; however, is thought to be inactive because it lacks several residues required for activity. This protein plays a critical role in tumor necrosis factor (TNF)-induced necroptosis, a programmed cell death process, via interaction with receptor-interacting protein 3 (RIP3), which is a key signaling molecule in necroptosis pathway. Inhibitor studies and knockdown of this gene inhibited TNF-induced necrosis. High levels of this protein and RIP3 are associated with inflammatory bowel disease in children. Alternatively spliced transcript variants have been described for this gene. [provided by RefSeq, Sep 2015]

Application

WB

Western blot analysis of Phospho-MLKL (S358) on different lysates with Rabbit anti-Phospho-MLKL (S358) antibody at 1/5000 dilution. Lane 1: HT-29 cell lysate, Lane 2: HT-29 treated with 20μM Z-VAD for 7.5 hours add 20ng/mL hTNF alpha and 100nM SM-164 for last 7 hours cell lysate, Lysates/proteins at 20 µg/Lane. Exposure time: 1 minute 50 seconds; 4-20% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody at 1/5000 dilution was used in primary antibody dilution at 4℃ overnight. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1/50000 dilution was used for 1 hour at room temperature.

DB

Dot blot analysis of Phospho-MLKL (S358) on different peptides with Rabbit anti-Phospho-MLKL (S358) antibody at 1/5000 dilution. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1/50000 dilution for 1 hour at room temperature. Lane 1: Unmodified MLKL peptide (negative), Lane 2: Phospho-MLKL (S358) peptide (positive), Proteins loading: 100ng, 25ng, 5ng; Blocking and dilution buffer: 5% NFDM/TBST; Exposure time: 5 seconds.
