Gene Summary Histones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. This structure consists of approximately 146 bp of DNA wrapped around a nucleosome, an octamer composed of pairs of each of the four core histones (H2A, H2B, H3, and H4). The chromatin fiber is further compacted through the interaction of a linker histone, H1, with the DNA between the nucleosomes to form higher order chromatin structures. This gene is intronless and encodes a replication-dependent histone that is a member of the histone H3 family. Transcripts from this gene lack polyA tails; instead, they contain a palindromic termination element. This gene is found in the large histone gene cluster on chromosome 6p22-p21.3. Western blot analysis using Histone H3(citrulline R2/R8/R17) antibody against Lane 1:Untreated HEK-293T (human embryonic kidney) transfected with PADI4 expression vector containing a myc-His-tag, whole cell lysate at 20 µg.Lane 2:HEK-293T transfected with PADI4 expression vector containing a myc-His-tag treated with 10 mM calcium chloride and 10 μM lonomycin for 4 hours, whole cell lysate at 20 µg.Lane 3:Untreated NIH/3T3 (mouse embryonic fibroblast) transfected with PADI4 expression vector containing a myc-His-tag, whole cell lysate at 20 µg.Lane 4:NIH/3T3 transfected with PADI4 expression vector containing a myc-His-tag treated with 10 mM calcium chloride and 10 μM lonomycin for 4 hours, whole cell lysate at 20 µg. 12% SDS-PAGE gel.Sample loading: 20μg /lane. Transfer the proteins onto a PVDF membrane (OM790003), and block it with TBST (OM750016) plus skimmed milk powder(OM750084) for one hour. Dilute the primary antibody with the antibody diluent (OM750012) at a ratio of 1:2000, and incubate it overnight at 4°C. Wash the membrane three times with TBST (OM750016), 5 minutes each time. At room temperature, dilute the secondary antibody, Goat Anti-Rabbit IgG(H&L)-HRP (OM643487), at a ratio of 1:20000 and incubate for one hour. Wash the membrane three times with TBST (OM750016) again, 5 minutes each time. Use ECL (OM625701) for luminescence.staining time: 60S.
