HEPES (N-2-Hydroxyethylpiperazine-N'-2-ethanesulfonic acid) is a zwitterionic organic chemical buffer that is commonly used in cell culture media. When it is necessary to handle cell cultures outside a CO2 incubator for an extended period, adding 10–25 mM HEPES can provide additional buffering capacity. HEPES has advantages such as being impermeable to cell membranes, having a limited impact on biochemical reactions, possessing chemical and enzymatic stability, and exhibiting extremely low absorbance in the visible light and ultraviolet light ranges. Therefore, it is an excellent buffering choice for many cell culture systems. |
