Insulin-Transferrin-Selenium (ITS-G) is used as a basal medium supplement to reduce the amount of fetal bovine serum (FBS) required for culturing cells. The ITS-G solution was initially used in combination with RPMI 1640 and Minimum Essential Medium (MEM), and is now commonly used with other basal media to support a variety of cell types in the presence of 2–4% FBS. Insulin promotes the uptake of glucose and amino acids, adipogenesis, intracellular transport, and the synthesis of proteins and nucleic acids. Transferrin is an iron carrier and also helps to reduce the toxic levels of oxygen free radicals and peroxides. Selenium (provided as sodium selenite) is a cofactor for glutathione peroxidase and other proteins and is used as an antioxidant in the medium. |
