Product Profile
Product Name CaMKII Antibody (M-176)
Antibody Type Primary Antibodies
Antigen Alias epitope corresponding to amino acids 303-478 mapping at the C-terminus of CAMKIIα of mouse origin
Modification Notes The Ca2+/calmodulin-dependent protein kinases (CaM kinases) comprise a structurally related subfamily of serine/threonine kinases which include CaMKI, CaMKII and CaMKIV. CaMKII is a ubiquitously expressed serine/threonine protein kinase that is activated by Ca2+and calmodulin (CaM) and has been implicated in regulation of the cell cycle and transcription. There are four CaMKII isozymes designated å, ∫, © and ∂, which may or may not be co-expressed in the same tissue type. CaMKIV is stimulated by Ca2+ and CaM but also requires phosphorylation by a CaMK for full activation. Stimulation of the T cell receptor CD3 signaling complex with an anti-CD3 monoclonal antibody leads to a 10-40 fold increase in CaMKIV activity. An additional kinase, CaMKK, functions to activate CaMKI through the specific phosphorylation of the regulatory Threonine residue at position 177.

Application

Application

Immunoperoxidase staining of formalin fixed, paraffin-embedded human bronchus tissue showing cytoplasmic and nuclear staining of respiratory epithelial cells and glandular cells.

Application

Immunofluorescence staining of methanol-fixed HeLa cells showing cytoplasmic localization.

Application

Immunoperoxidase staining of formalin-fixed, paraffin-embedded human breast tumor showing cytoplasmic staining.

Application

Western blot analysis of CaMKII expression in rat brain extract.

Application

Blots were probed with CaMKII (M-176): (A) and p-CaMKIIα (Thr 286)-R: (B,C). In B and C, the antibody was preincubated with cognate non-phosphorylated or phosphorylated peptide, respectively.
Application Notes recommended for detection of all CaMKII subunits of mouse, rat and human origin by WB, IP, IF, IHC(P) and ELISA also reactive with additional species, including equine, canine, bovine, porcine and avian:
