Product Profile
Product Name involucrin Antibody (H-120)
Antibody Type Primary Antibodies
Conjugation Agarose
Modification Acetylated
Modification Notes Involucrin is a precursor protein of the keratinocyte cornified envelope, which is formed beneath the inner surface of the cell membrane during terminal differentiation. Involucrin first appears in the cell cytosol but ultimately becomes cross-linked to membrane proteins by transglutaminase. During keratinocyte terminal differentiation, glutamine residues of involucrin become covalently cross-linked to other envelope precursors via covalent epsilon-(gamma-glutamyl) lysine bonds. Moreover, its large size allows involucrin to cross-link molecules that are separated by substantial distances in the cornified envelope. These properties allow a single involucrin molecule to form multiple cross-links, in multiple spatial planes, with other envelope precursors. Involucrin is specifically expressed in Chinese hamster ovarian cells (fibroblasts), PtK2 rat kangaroo kidney cells (simple epithelial), and rat epidermal keratinocytes (stratifying squamous epithelial).

Application

Application

Immunoperoxidase staining of formalin fixed, paraffin-embedded human cervix tissue showing cytoplasmic staining of squamous epithelial cells.

Application

Western blot analysis of involucrin expression in CCD-1064Sk (A), SK-MEL-28 (B), A-431 (C), HT-1080 (D), A-375 (E) and SW480 (F) whole cell lysates.

Application

Immunofluorescence staining of methanol-fixed HeLa cells showing cytoplasmic localization.

WB

- miR-339-5p was a negative regulator in loureirin A-induced HFSC differentiation. K10 and involucrin expression decreased in the miR-339-5p mimic group compared with the mimic NC group, and increased in the miR-339-5p inhibitor group compared with the inhibitor NC group. *P<0.05 vs. the respective control groups. miR-339-5p, microRNA-339-5p; NC, negative control; K10, keratin, type I cytoskeletal 10.

WB

- Wnt3a, involucrin and K10 expression decreased in hair follicle stem cells transfected with siDLX5. *P<0.05 vs control group. K10, keratin, type I cytoskeletal 10; siDLX5, homeobox protein DLX-5 small interfering RNA; Wnt3a, protein Wnt-3a.

I F

CK5 and involucrin were red, while the nuclei were blue (a).

WB

The western blot results revealed that the expression of CK5 and involucrin was upregulated in the miR-203a-3p mimic group compared with the mimic NC group, while it was downregulated in the miR-203a-3p inhibitor group compared with the inhibitor NC group (b).

WB

Western blotting showed that the expression of CK5 and involucrin was upregulated after transfection of short interfering siSmad1-003.
