Product Profile
Product Name TFIIB Antibody (C-18)
Antibody Type Primary Antibodies
Antigen Alias epitope mapping near the C-terminus of TFIIB of human origin
Modification Notes In eukaryotic systems, initiation of transcription from protein-coding genes is a complex process requiring RNA polymerase II and broad families of auxiliary transcription factors. Such factors can be divided into two major functional classes: the basal factors that are required for transcription of all Pol II genes, including TFIIA, TFIIB, TFIID, TFIIE, TFIIF and TFIIH; and sequence-specific factors that regulate gene expression. The basal transcription factors and Pol II form a specific multiprotein complex near the transcription start site by interacting with core promotor elements such as the TATA box generally located 25-30 base pairs upstream of the transcription start site. Template commitment is established by the initial binding of TFIID to the “TATA” element of the promotor, a step which may be facilitated by TFIIA. TFIIB then acts as the bridge between TFIID and RNA polymerase II.

Application

Application

Western blot analysis of TFIIB expression in HeLa (A), Hep G2 (B) and U-937 (C) nuclear extracts.

Application

Western blot analysis of TFIIB expression in non-transfected: (A) and mouse TFIIB transfected: (B) 293T whole cell lysates.

Application

ChIP analysis of TFIIB recruitment to genomic amplicons. Two (1,2) different human genomic TBP and three control amplicons (3-5) were analyzed by quantitative PCR (primer sequences available in on-line supplemental data). Data generated in collaboration with Drs. N. Trinklein and R. Myers, Stanford University (ENCODE Project).

Application

Indirect, intracellular FCM analysis of fixed and permeabilized K-562 cells stained with TFIIB (C-18), followed by PE-conjugated goat anti-rabbit IgG: . Black line histogram represents the isotype control, normal rabbit IgG: .

Application

Immunofluorescence staining of methanol-fixed K-562 cells showing nuclear staining.

Application

Western blot analysis of TFIIB p33 expression in Jurkat (A) and K-562 (B) whole cell lysates.
