Key Feature
Clonality Polyclonal
Isotype Ig
Host Species Rabbit
Tested Applications WBIHCIF
WB:1:100~500 IHC
Species Reactivity Human
Concentration 1mg/ml
Target Information
Gene Synonyms LYSP100
Alternative Names SP140
LYSP100
Nuclear body protein SP140
Lymphoid-restricted homolog of Sp100
Nuclear autoantigen Sp-140
Speckled 140 kDa
Molecular Weight(MW) 98223 Da
Function Component of the nuclear body, also known as nuclear domain 10, PML oncogenic domain, and KR body. May be involved in the pathogenesis of acute promyelocytic leukemia and viral infection
Tissue Specificity High levels in spleen and peripheral blood leukocytes, much lower levels in thymus, prostate, ovary, small intestine, and colon. Very low levels in heart, brain, placenta, lung, liver, skeletal muscle, kidney, and pancreas
Cellular Localization Nucleus. Cytoplasm. Note=Localized to nuclear structures termed LANDS, for LYSp100-associated nuclear domains. LANDS are globular, electron-dense structures most often found in the nucleoplasm, but also found at the nuclear membrane and in the cytoplasm, suggesting that these structures may traffic between the cytoplasm and the nucleus

Application

Application

SP140 Antibody (Center) western blot analysis in HL-60 cell line lysates (35ug/lane).This demonstrates the SP140 antibody detected the SP140 protein (arrow).

Application

SP140 Antibody (Center) immunohistochemistry analysis in formalin fixed and paraffin embedded human kidney tissue followed by peroxidase conjugation of the secondary antibody and DAB staining. This data demonstrates the use of SP140 Antibody (Center) for immunohistochemistry. Clinical relevance has not been evaluated.

Application

Fluorescent confocal image of Hela cell stained with SP140 Antibody (Center)(Cat#AP12872c).Hela cells were fixed with 4% PFA (20 min), permeabilized with Triton X-100 (0.1%, 10 min), then incubated with SP140 primary antibody (1:25, 1 h at 37℃). For secondary antibody, Alexa Fluor® 488 conjugated donkey anti-rabbit antibody (green) was used (1:400, 50 min at 37℃).Cytoplasmic actin was counterstained with Alexa Fluor® 555 (red) conjugated Phalloidin (7units/ml, 1 h at 37℃). Nuclei were counterstained with DAPI (blue) (10 µg/ml, 10 min). SP140 immunoreactivity is localized to Cytoplasm and Nucleus significantly.
Application Notes WB:1:100~500 IHC
