Target Information
Alternative Names BUB3
Mitotic checkpoint protein BUB3
Molecular Weight(MW) 37155 Da
Function Has a dual function in spindle-assembly checkpoint signaling and in promoting the establishment of correct kinetochore-microtubule (K-MT) attachments. Promotes the formation of stable end-on bipolar attachments. Necessary for kinetochore localization of BUB1. Regulates chromosome segregation during oocyte meiosis. The BUB1/BUB3 complex plays a role in the inhibition of anaphase-promoting complex or cyclosome (APC/C) when spindle-assembly checkpoint is activated and inhibits the ubiquitin ligase activity of APC/C by phosphorylating its activator CDC20. This complex can also phosphorylate MAD1L1
Tissue Specificity This BUB3 antibody is generated from rabbits immunized with a KLH conjugated synthetic peptide between 299~328 amino acids from the C-terminal region of human BUB3.
Cellular Localization Nucleus. Chromosome, centromere, kinetochore (By similarity). Note=Starts to localize at kinetochores in prometaphase I (Pro-MI) stage and maintains the localization until the metaphase I-anaphase I (MI-AI) transition (By similarity)

Application

Application

Western blot analysis of BUB3 Antibody (C-term) in Hela cell line lysates (35ug/lane). BUB3 (arrow) was detected using the purified Pab.

Application

BUB3 Antibody (C-term) IHC analysis in formalin fixed and paraffin embedded testis tissue followed by peroxidase conjugation of the secondary antibody and DAB staining. This data demonstrates the use of the BUB3 Antibody (C-term) for immunohistochemistry. Clinical relevance has not been evaluated.

Application

BUB3 Antibody (C-term) flow cytometric analysis of Hela cells (right histogram) compared to a negative control cell (left histogram).FITC-conjugated goat-anti-rabbit secondary antibodies were used for the analysis.

Application

Western blot analysis of BUB3 (arrow) using rabbit polyclonal BUB3 Antibody (C-term) . 293 cell lysates (2 ug/lane) either nontransfected (Lane 1) or transiently transfected (Lane 2) with the BUB3 gene.
Application Notes WB:1:100~500 IHC
