Function Probably has a broad role in mammalian development as a repressor of developmentally regulated gene expression. May act by preventing binding of positively-activing CCAAT factors to promoters. Component of nf-munr repressor; binds to the matrix attachment regions (MARs) (5' and 3') of the immunoglobulin heavy chain enhancer. Represses T-cell receptor (TCR) beta enhancer function by binding to MARbeta, an ATC-rich DNA sequence located upstream of the TCR beta enhancer (By similarity)
Tissue Specificity This CUX1 antibody is generated from rabbits immunized with a KLH conjugated synthetic peptide between 1354-1382 amino acids from the C-terminal region of human CUX1.
Cellular Localization Nucleus.

Application

Application

CUX1 Antibody (C-term) western blot analysis in MCF-7 cell line lysates (35ug/lane).This demonstrates the CUX1 antibody detected the CUX1 protein (arrow).

Application

CUX1 Antibody (C-term) immunohistochemistry analysis in formalin fixed and paraffin embedded human kidney tissue followed by peroxidase conjugation of the secondary antibody and DAB staining. This data demonstrates the use of the CUX1 Antibody (C-term) for immunohistochemistry. Clinical relevance has not been evaluated.

Application

Fluorescent confocal image of Hela cell stained with CUX1 Antibody (C-term)(Cat#AP5895b).Hela cells were fixed with 4% PFA (20 min), permeabilized with Triton X-100 (0.1%, 10 min), then incubated with CUX1 primary antibody (1:25, 1 h at 37℃). For secondary antibody, Alexa Fluor® 488 conjugated donkey anti-rabbit antibody (green) was used (1:400, 50 min at 37℃).Cytoplasmic actin was counterstained with Alexa Fluor® 555 (red) conjugated Phalloidin (7units/ml, 1 h at 37℃). Nuclei were counterstained with DAPI (blue) (10 µg/ml, 10 min). CUX1 immunoreactivity is localized to Nucleus significantly.
Application Notes WB:1:100~500 IHC
