Western blot analysis of Ser1981-phosphorylated ATM in gamma-irradiated HeLa cells.Application
Immunofluorescent analysis of Ser1981-phosphorylated ATM in control (left panel) or 100 mJ UV-stimulated 293 cells (right panel). Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes at room temperature and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with ATM (phospho Ser1981) antibody [10H11] at a dilution of 1:50 and incubated overnight in a humidified chamber. Cells were washed with PBST and incubated with a proper secondary antibody. Nuclei (blue) were stained with DAPI. | Product description | Mouse monoclonal antibody [10H11] to S1981-phosphorylated ATMThe protein encoded by this gene belongs to the PI3/PI4-kinase family. This protein is an important cell cycle checkpoint kinase that phosphorylates; thus, it functions as a regulator of a wide variety of downstream proteins, including tumor suppressor proteins p53 and BRCA1, checkpoint kinase CHK2, checkpoint proteins RAD17 and RAD9, and DNA repair protein NBS1. This protein and the closely related kinase ATR are thought to be master controllers of cell cycle checkpoint signaling pathways that are required for cell response to DNA damage and for genome stability.
