Product Profile
Product Name Fas antibody
Antibody Type Primary Antibodies
Product description Mouse Monoclonal antibody [LT95] to FasCD95, also known as FAS or APO-1, is a 36 kDa cell surface type I- membrane glycoprotein with an apparent molecular weight of 44 kDa on SDS-PAGE. CD95 is a member of the TNF receptor family, which includes TNFR-1, TNFR-2, CD27, CD30 and CD40. Binding of CD95 Ligand to CD95 or crosslinking of CD95 by anti-CD95 monoclonal antibodies leads to apoptosis of CD95 expressing cells. CD95 belongs to a subgroup of family members that have a death domain (DD) which contains 70 amino acids near the carboxyl-terminal region of the molecule. The binding of adaptor molecules to this DD is responsible for transmitting the death signal for apoptosis. Stimulation of CD95 results in aggregation of its DD, leading to the recruitment of FADD and caspase-8 that together with the receptor form the death-inducing signaling complex (DISC).
Immunogen Tissue / cell preparation (Human). (T cell lymphoma cells HUT-78).

Application

Application

Flow cytometry analysis of Fas (CD95) expression on the surface of various hematopoietic cell lines with purified anti-human CD95 (detection by Goat anti-mouse IgG1 PE). Histograms: Red – Isotype mouse IgG1 control staining Black – standard anti-CD95 monoclonal antibody (DX2) Green – anti-CD95 (LT95) A – JURKAT human peripheral blood T cell leukemia cell line B – RAMOS human Burkitt lymphoma cell line C – CEM human leukemia cell line D – MOLT-4 human acute lymphoblastic T cell leukemia cell line
Positive Control Reacts strongly with IM9 and HUT-78 cell lines. Reacts moderately with Raji, Ramos, U266, Jurkat, BJAB and ML-1 cell lines. Reacts weakly with KG1a, U937, KM3 and TF-1 cell lines. Does not react with K562 and THP-1 cell lines.
Application Notes FACS: Use at an assay dependant concentration. Does not induce Fas mediated apoptosis in susceptible cells. Optimal dilutions/concentrations should be determined by the end user.:
