| Product description | Mouse monoclonal to b - ARK1The s-adrenergic receptor kinase 1 (sARK1) was originally isolated as a ser/thr kinase that phosphorylates cardiac s-adrenergic receptors. It has subsequently been found to phosphorylate many other kinds of G protein coupled receptors (GPCR), as well. It is now recognized as one of a family of G protein coupled receptor kinases that are important in regulating the activity of GPCRs. Phosphorylation of GCPRs by sARK1, and other related kinases, generally occurs only after the receptor has been activated by ligand binding. The phosphorylation results in the uncoupling of the GPCR from G proteins by binding to s-arrestin. The binding of s-arrestin may also result in receptor internalization, mediated by clatherin and dynamin and/or the activation of the MAP kinase cascade. ßARK1 is found in many cell types and may also have a role in the regulation of the microtubular cytoskeleton. The levels of ßARK1 enzymatic activity, protein and mRNA are all elevated in a variety of human heart disease states, and the effect of this may contribute to the reduced effectiveness of the ß-adrenergic system in these disease states. |
