Product Profile
Product Name HDAC7 antibody
Antibody Type Primary Antibodies
Product description Rabbit polyclonal to HDAC7 - ChIP GradeHDAC7 is a member of the class II mammalian histone deacetylases, which plays an important role in modulating the eukaryotic chromatin structure. Human HDAC7 is composed of 912 amino acid residues. Although HDAC7 is localized mostly to the cell nucleus, it is also found in the cytoplasm, suggesting nucleo-cytoplasmic shuttling. The histone deacetylase activity of HDAC7 maps to a carboxy-terminal domain and is dependent on interaction with class I HDACs in the nucleus. It is an active component of different transcriptional corepressor complexes that can be recruited to specific promoter regions via interactions with a growing number of sequence specific transcriptional factors. HDAC7 catalyzes removal of acetyl-groups from acetyl-lysines of histones and promotes compaction of chromatin in these regions, leading to the inhibition of gene transcription.

Target HDAC7
Tissue Specificity The antibody detects ~105 kDa histone deacetylase 7. It does not cross-react with other HDAC proteins.
Cellular Localization Nuclear and cytoplasmic
Application

Application

Western blot analysis of HDAC-7 expresssion in mouse small intestine tissue lysate.

Application

Sonicated Chromatin prepared from untreated (UI) or 17beta-estradiol (E2) treated MCF7 cells was subjected to the ChIP procedure with to HDAC7 and the immunoprecipitated chromatin was analysed in the proximal region of the estrogen-responsive pS2 promoter (as shown above) and quantified by real-time PCR (values are nomalized over inputs). The primers are designed to follow the nucleosome E (including the Estrogen Responsive Element ERE). 10 ul of and 2x10*6cells were used in each ChIP experiment.
Application Notes ChIP: Use 10μl for 10^6 cells. IHC-Fr: Use at a concentration of 30 μg/ml. WB: Use at a concentration of 1 μg/ml. Predicted molecular weight: 102.9 kDa. Optimal dilutions/concentrations should be determined by the end user.
