Product Profile
Product Name Involucrin antibody
Antibody Type Primary Antibodies
Product description Mouse monoclonal [0.N.341] to InvolucrinInvolucrin, a component of the keratinocyte crosslinked envelope, is found in the cytoplasm and crosslinked to membrane proteins by transglutaminase. Involucrin is expressed in a range of stratified squamous epithelia, including the cornea. In normal epidermis, it is first expressed in the upper spinous layers, and in keratinocyte cultures it is expressed by all cells that have left the basal layer. Involucrin expression is abnormal in squamous cell carcinomas and premalignant lesions and is reduced in severe dysplasias of the larynx and cervix. Its gene is mapped to 1q21, among calpactin I light chain, trichohyalin, profillaggrin, loricrin, and calcyclin.
Immunogen Full length native protein (purified) (Human).
Key Feature
Clonality Monoclonal
Isotype IgG1
Clone Number 0.N.341
Host Species Mouse
Tested Applications ELISAICC/IFIHC-PIPWB
ELISA: Use at an assay dependent dilution. IF: Use at an assay dependent dilution. IHC-P: Use at a concentration of 1 - 2 ?g/ml. Perform enzymatic antigen retrieval before commencing with IHC staining protocol. For staining formalin-fixed tissues, digest sections with trypsin at 1mg/ml PBS, for 10 min at 37?C. Note: Enzyme digestion is better than citrate buffer based epitope unmasking. IP: Use 2 ?g/mg protein lysate (Denatured verified Use Protein G). WB: Use at a concentration of 1 - 2 ?g/ml. (Human form at 170 kDa see under """"Specificity"""" for other species molecular weights.) Optimal dilutions/concentrations should be determined by the end user.:

Tissue Specificity Recognizes involucrin at 66-170kD, depending on cell type. Stains involucrin in a variety of sizes: 170kD in MCF7 cells, a doublet of ~115kD and 150kD in gorilla and owl monkey, 66kD in canine and a doublet of 105kD in porcine.
Cellular Localization Cytoplasmic
Positive Control LS174T cells and skin
