Product Profile
Product Name p27 Kip1 (phospho Thr187) antibody
Antibody Type Primary Antibodies
Product description Rabbit Polyclonal antibody to p27 Kip1 (phospho Thr187)This gene encodes a cyclin-dependent kinase inhibitor, which shares a limited similarity with CDK inhibitor CDKN1A/p21. The encoded protein binds to and prevents the activation of cyclin E-CDK2 or cyclin D-CDK4 complexes, and thus controls the cell cycle progression at G1. The degradation of this protein, which is triggered by its CDK dependent phosphorylation and subsequent ubiquitination by SCF complexes, is required for the cellular transition from quiescence to the proliferative state. Mutations in this gene are associated with multiple endocrine neoplasia type IV (MEN4). [provided by RefSeq, Apr 2014]

Application

Application

Western blot analysis of p27 Kip1 (pT187) expression in HeLa (A); MCF7 (B) whole cell lysates.

Application

Immunohistochemical analysis of p27 Kip1 (pT187) staining in human breast cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugad compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX. w

Application

Immunofluorescent analysis of p27 Kip1 (pT187) staining in A431 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 i??i??C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue).
Application Notes WB: 1:500 - 1:1000. IHC-P: 1:100 - 1:200. ICC/IF: 1:100 - 1:500
