Product Profile
Product Name PKA alpha + beta (phospho Thr197) antibody
Antibody Type Primary Antibodies
Product description Rabbit polyclonal to PKA alpha + beta ( catalytic subunits ) ( phospho T197 )c-AMP-dependent Protein Kinase (PKA) is a serine/threonine kinase that regulates a number of cellular processes including proliferation, ion transport and gene transcription. PKA is composed of conserved catalytic subunits and regulatory subunits that dissociate upon activation by cAMP. The catalytic subunit of PKA contains the activation loop and mediates DNA binding and substrate recognition. The catalytic subunit is assembled and expressed as an active form and is phosphorylated on threonine 197 by PDK-1 in the activation loop. Phosphorylation of threonine 197 is critical for PKA maturation, optimal conformation and catalytic activity and plays a key role in mediating its biological functions.
Immunogen Synthetic phosphopeptide derived from a region of human PKA that contains threonine 197.
Modification p-Thr197

Application

Application

Lysates prepared from Y1 Adrenocortical cell lysates untreated (1-4) or treated with l phosphatase (5), then incubated with anti-PKA [pT197] after prior incubation with: no peptide (1, 5), the non-phosphopeptide (2), a generic [pT] peptide (3), or, the phosphopeptide immunogen (4). Data show antibody phospho-site specificity by peptide competition, and that phosphatase stripping eliminates the signal, as expected.
Application Notes WB: Use at a concentration of 0.1 - 0.75 μg/ml. Detects a band of approximately 42 kDa. Optimal dilutions/concentrations should be determined by the end user.
