Product Profile
Product Name TNF-a Antibody
Antibody Type Primary Antibodies
Antigen Alias DIF, TNFA, TNFSF2, TNF-alpha, Tumor necrosis factor, Cachectin, TNF-a
Product description Tumor Necrosis Factor Alpha (TNF alpha) is a protein secreted by lipopolysaccharide stimulated macrophages, and causes tumor necrosis when injected into tumour bearing mice. TNF alpha is believed to mediate pathogenic shock and tissue injury associated with endotoxemia.
Immunogen Produced from sera of rabbits pre-immunized with highly pure (>98%) recombinant hTNF-alpha (human Tumor Necrosis Factor-alpha).
Key Feature
Clonality Polyclonal
Host Species Rabbit
Neutralization: To yield one-half maximal inhibition [ND50] of the biological activity of hTNF-α (0.25 ng/mL), a concentration of 0.012 - 0.014 μg/mL of this antibody is required. ELISA: To detect hTNF-α by direct ELISA (using 100 μL/well antibody solution) a concentration of at least 0.5 μg/mL of this antibody is required. This antigen affinity purified antibody, in conjunction with compatible secondary reagents, allows the detection of 0.2 - 0.4 ng/well of recombinant hTNF-α. Sandwich: To detect hTNF-α by sandwich ELISA (using 100 μL/well antibody solution) a concentration of 0.5 - 2.0 μg/mL of this antibody is required. This antigen affinity purified antibody, in conjunction with our Biotinylated Anti-Human TNF-α (XP-5282Bt) as a detection antibody, allows the detection of at least 0.2 - 0.4 ng/well of recombinant hTNF-α. Western Blot: To detect hTNF-α by Western Blot analysis this antibody can be used at a concentration of 0.1 - 0.2 μg/mL. Used in conjunction with compatible secondary reagents the detection limit for recombinant hTNF-α is 1.5 - 3.0 ng/lane, under either reducing or non-reducing conditions.:

Application

Application

To detect Human TNF-alpha by sandwich ELISA (using 100 ul/well antibody solution) a concentration of 0.5 - 2.0 ug/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with ProSci’s Biotinylated Anti-Human TNF-alpha (XP-5282Bt) as a detection antibody, allows the detection of at least 0.2 - 0.4 ng/well of recombinant Human TNF-alpha.

Application

This antibody stained formalin-fixed, paraffin-embedded sections of human breast invasive ductal carcinoma. The recommended concentrations are 0.5 ug/ml-1.0 ug/ml with an overnight incubation at 4˚C. An HRP-labeled polymer detection system was used with a DAB chromogen. Heat induced antigen retrieval with a pH 6.0 Sodium Citrate buffer is recommended. Optimal concentrations and conditions may
