Product Profile
Product Name Fibrinogen Rabbit Polyclonal Antibody
Antibody Type Primary Antibodies
Product description

The plasma glycoprotein Fibrinogen is synthesized in the liver and comprises three structurally different subunits: a, b and g. Fibrinogen is important in platelet aggregation, the final step of the coagulation cascade (i.e. the formation of Fibrin) and determination of plasma viscosity and erythrocyte aggregation. It is both constitutively expressed and inducible during an acute phase reaction. Hemostasis following tissue injury deploys essential plasma procoagulants (Prothrombin and Factors X, IX, V and VIII), which are involved in a blood coagulation cascade leading to the formation of insoluble Fibrin clots and the promotion of platelet aggregation. Following vascular injury, Fibrinogen is cleaved by Thrombin to form Fibrin, which is the most abundant component of blood clots. The cleavage products of Fibrinogen regulate cell adhesion and spreading, display vasoconstrictor and chemotactic activities, and are mitogens for several cell types.

Application

WB

Western blot analysis of Fibrinogen on different lysates with Rabbit anti-Fibrinogen antibody at 1/5,000 dilution. Lane 1: Hep G2 (Human hepatocellular carcinoma cell) cell lysate, Lane 2: Human plasma lysate, Lysates/proteins at 10 µg/Lane. Exposure time: 4 seconds; Blocking: 5% NFDM/TBST, 1 hour at room temperature, Primary antibody: 1/5,000 in primary antibody dilution buffer, overnight at 4 ℃ Secondary antibody: Goat anti-Rabbit IgG-HRP, 1/50,000 in 5% NFDM/TBST, 1 hour at room temperature.

ICC/IF

ICC staining Fibrinogen in NIH-3T3 cells (green). The nuclear counter stain is DAPI (blue). Cells were fixed in paraformaldehyde, permeabilised with 0.25% Triton X100/PBS.

ICC/IF

ICC staining Fibrinogen in A549 cells (green). The nuclear counter stain is DAPI (blue). Cells were fixed in paraformaldehyde, permeabilised with 0.25% Triton X100/PBS.

IHC

Immunohistochemical analysis of paraffin-embedded human placenta tissue with Rabbit anti-Fibrinogen antibody at 1/4,000 dilution. The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 20 minutes. The tissues were blocked in 1% BSA for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody at 1/8,000 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

IHC

Immunohistochemical analysis of paraffin-embedded human lung tissue with Rabbit anti-Fibrinogen antibody at 1/4,000 dilution.
