Western blot analysis of RAB7 on different lysates with Rabbit anti-RAB7 antibody at 1/5,000 dilution. Lane 1: HeLa cell lysate, Lane 2: MCF7 cell lysate, Lane 3: Neuro-2a cell lysate, Lane 4: C2C12 cell lysate, Lane 5: PC-12 cell lysate, Lane 6: C6 cell lysate, Lysates/proteins at 15 µg/Lane. Exposure time: 17 seconds; 4-20% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody at 1/5,000 dilution was used in 5% NFDM/TBST at 4℃ overnight. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1/50,000 dilution was used for 1 hour at room temperature.IHC

The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 20 minutes. The tissues were blocked in 1% BSA for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody at 1/1,000 dilution for 1 hour at room temperature. | Product description | The Ras-related superfamily of guanine nucleotide binding proteins, which includes the Ral/Rec, Rap, R-Ras, and Rho/Rab subfamilies, exhibit 30-60% homology with Ras p21. Accumulating data suggests an important role for Rab proteins, either in endocytosis or in biosynthetic protein transport. The transport of newly synthesized proteins from the endoplasmic reticulum to various stacks of the Golgi complex and to secretory vesicles involves at each stage the movement of carrier vesicles, a process that appears to involve Rab protein function. The possibility that Rab proteins might also direct the exocytosis from secretory vesicles to the plasma membrane is supported by the observation that in yeast, the Sec4 protein, which is 40% homologous to Rab proteins, is associated with secretory vesicles.
