Western blot analysis of SMC1A on different lysates with Rabbit anti-SMC1A antibody at 1/2,000 dilution. Lane 1: HeLa cell lysate, Lane 2: A431 cell lysate, Lane 3: 293T cell lysate, Lane 4: NIH/3T3 cell lysate, Lane 5: 4T1 cell lysate, Lane 6: PC-12 cell lysate, Lysates/proteins at 20 µg/Lane. Exposure time: 3 minutes; 4-20% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody at 1/2,000 dilution was used in 5% NFDM/TBST at 4℃ overnight. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1/50,000 dilution was used for 1 hour at room temperature.IHC

Immunocytochemistry analysis of PC-12 cells labeling SMC1A with Rabbit anti-SMC1A antibody at 1/100 dilution. Cells were fixed in 100% precooled methanol for 5 minutes at room temperature, then blocked with 1% BSA in 10% negative goat serum for 1 hour at room temperature. Cells were then incubated with Rabbit anti-SMC1A antibody at 1/100 dilution in 1% BSA in PBST overnight at 4 ℃. Goat Anti-Rabbit IgG H&L (488) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. Nuclear DNA was labelled in blue with DAPI. Beta tubulin (red) was stained at 1/100 dilution overnight at +4℃. | Product description | The SMC (structural maintenance of chromosomes) family of proteins form heterodimeric complexes that modulate sister chromatid cohesion and chromosome condensation for mitosis. SMC1α (structural maintenance of chromosomes protein 1A), also known as SMC1, SMCB, CDLS2, SB1.8, SMC1L1 or DXS423E, is a 1,233 amino acid nuclear protein that is involved in chromosome cohesion during the cell cycle. SMC1α interacts with BRCA1 and is phosphorylated by ATM, indicating a potential role in DNA repair. SMC1α is a component of the cohesion complex, which is required for the cohesion of sister chromatids after DNA replication. Mutations in the gene encoding SMC1α may be the cause of Cornelia de Lange syndrome (CdLS), which is a clinically heterogeneous developmental disorder characterized by facial dysmorphia, upper limb malformations, growth and cognitive retardation. |
