The sample was formaldehyde fixed and a heat mediated antigen retrieval step in citrate buffer was performed. The sample was then blocked and incubated with the primary antibody at 4°C overnight. COL22A1 Antibody staining Hela cells by ICC/IF. The samples were fixed with PFA and permeabilized in 0.1% Triton X-100, then blocked in 10% serum for 45 minutes at 25°C. Samples were then incubated with primary Ab and mouse anti-beta tubulin Ab for 1 hour at 37°C. An AlexaFluor594 conjugated goat anti-rabbit IgG Ab(Red) and an AlexaFluor488 conjugated goat anti-mouse IgG Ab(Green) were used as the secondary antibody. | Gene Summary | This gene encodes member of the collagen family which is thought to contribute to the stabilization of myotendinous junctions and strengthen skeletal muscle attachments during contractile activity. It belongs to the fibril-associated collagens with interrupted triple helix (FACIT) subset of the collagen superfamily, which associate with collagen fibers through their C-terminal collagenous domains and mediate protein-protein interactions through their N-terminal noncollagenous domains. The encoded protein is deposited in the basement membrane zone of the myotendinous junction which is present only at the tissue junctions of muscles, tendons, the heart, articular cartilage, and skin.
