Target Information
Gene Symbol DUSP2
Gene Synonyms PAC1
PAC-1
Gene Full Name dual specificity phosphatase 2
Gene Summary The protein encoded by this gene is a member of the dual specificity protein phosphatase subfamily. These phosphatases inactivate their target kinases by dephosphorylating both the phosphoserine/threonine and phosphotyrosine residues. They negatively regulate members of the mitogen-activated protein (MAP) kinase superfamily (MAPK/ERK, SAPK/JNK, p38), which are associated with cellular proliferation and differentiation. Different members of the family of dual specificity phosphatases show distinct substrate specificities for various MAP kinases, different tissue distribution and subcellular localization, and different modes of inducibility of their expression by extracellular stimuli. This gene product inactivates ERK1 and ERK2, is predominantly expressed in hematopoietic tissues, and is localized in the nucleus. [provided by RefSeq, Jul 2008]

Application

WB

Western blot analysis of DUSP2 using anti-DUSP2 antibody. The sample well of each lane was loaded with 30 ug of sample under reducing conditions. After electrophoresis, proteins were transferred to a membrane. Then the membrane was incubated with rabbit anti-DUSP2 antigen affinity purified polyclonal antibody at a dilution of 1:1000 and probed with a goat anti-rabbit IgG-HRP secondary antibody. The signal is developed using ECL Plus Western Blotting Substrate.

IHC

IHC analysis of DUSP2 using anti-DUSP2 antibody. DUSP2 was detected in a paraffin-embedded section of human tonsil tissue. The tissue section was incubated with rabbit anti-DUSP2 Antibody at a dilution of 1:200 and developed using HRP Conjugated Rabbit IgG Super Vision Assay Kit with DAB as the chromogen.

ICC/IF

ICC/IF analysis of DUSP2 using anti-DUSP2 antibody and anti-Alpha Tubulin antibody. DUSP2 was detected in an immunocytochemical section of Hela cells. The section was incubated with rabbit anti-DUSP2 Antibody at a dilution of 1:100. Fluoro488-conjugated Anti-rabbit IgG Secondary Antibody (green) and Cy3-conjugated Anti-mouse IgG Secondary Antibody (red) were used as secondary antibody.

FC

Flow Cytometry analysis of RT4 cells using anti-DUSP2 antibody. Overlay histogram showing RT4 cells stained with anti-DUSP2 antibody (Blue line). To facilitate intracellular staining, cells were fixed with 4% paraformaldehyde and permeabilized with permeabilization buffer. The cells were blocked with 10% normal goat serum. And then incubated with rabbit anti-DUSP2 Antibody at 1:100 dilution for 30 min at 20°C. Fluoro488 conjugated goat anti-rabbit IgG was used as secondary antibody at 1:100 dilution for 30 minutes at 20°C. Isotype control antibody (Green line) was rabbit IgG at 1:100 dilution used under the same conditions. Unlabelled sample without incubation with primary antibody and secondary antibody (Red line) was used as a blank control.
