Gene Full Name poly(ADP-ribose) polymerase 1
Gene Summary This gene encodes a chromatin-associated enzyme, poly(ADP-ribosyl)transferase, which modifies various nuclear proteins by poly(ADP-ribosyl)ation. The modification is dependent on DNA and is involved in the regulation of various important cellular processes such as differentiation, proliferation, and tumor transformation and also in the regulation of the molecular events involved in the recovery of cell from DNA damage. In addition, this enzyme may be the site of mutation in Fanconi anemia, and may participate in the pathophysiology of type I diabetes. [provided by RefSeq, Jul 2008]

Application

WB

Western blot analysis of PARP1 using anti-PARP1 antibody. The sample well of each lane was loaded with 30 ug of sample under reducing conditions. After electrophoresis, proteins were transferred to a membrane. Then the membrane was incubated with mouse anti-PARP1 antigen affinity purified monoclonal antibody at a dilution of 1:1000 and probed with a goat anti-mouse IgG-HRP secondary antibody. The signal is developed using ECL Plus Western Blotting Substrate.

IHC

IHC analysis of PARP1 using anti-PARP1 antibody. PARP1 was detected in a paraffin-embedded section of human gallbladder adenocarcinoma tissue. Biotinylated goat anti-mouse IgG was used as secondary antibody. The tissue section was incubated with mouse anti-PARP1 Antibody at a dilution of 1:200 and developed using Strepavidin-Biotin-Complex (SABC) with DAB as the chromogen.

IHC

IHC analysis of PARP1 using anti-PARP1 antibody. PARP1 was detected in a paraffin-embedded section of human ovarian serous adenocarcinoma tissue. Biotinylated goat anti-mouse IgG was used as secondary antibody. The tissue section was incubated with mouse anti-PARP1 Antibody at a dilution of 1:200 and developed using Strepavidin-Biotin-Complex (SABC)with DAB as the chromogen.

ICC/IF

ICC/IF analysis of PARP1 using anti-PARP1 antibody. PARP1 was detected in an immunocytochemical section of A431 cells. The section was incubated with mouse anti-PARP1 Antibody at a dilution of 1:100. Fluoro488-conjugated Anti-mouse IgG Secondary Antibody (green) was used as secondary antibody. The section was counterstained with DAPI (Blue).

FC

Flow Cytometry analysis of HL-60 cells using anti-PARP1 antibody. Overlay histogram showing HL-60 cells stained with anti-PARP1 antibody (Blue line). To facilitate intracellular staining, cells were fixed with 4% paraformaldehyde and permeabilized with permeabilization buffer. The cells were blocked with 10% normal goat serum. And then incubated with mouse anti-PARP1 Antibody at 1:100 dilution for 30 min at 20°C. Fluoro488 conjugated goat anti-mouse IgG was used as secondary antibody at 1:100 dilution for 30 minutes at 20°C. Isotype control antibody (Green line) was mouse IgG at 1:100 dilution used under the same conditions. Unlabelled sample without incubation with primary antibody and secondary antibody (Red line) was used as a blank control.
