Target Information
Gene Symbol PIR
Gene Full Name pirin
Gene Summary This gene encodes a member of the cupin superfamily. The encoded protein is an Fe(II)-containing nuclear protein expressed in all tissues of the body and concentrated within dot-like subnuclear structures. Interactions with nuclear factor I/CCAAT box transcription factor as well as B cell lymphoma 3-encoded oncoprotein suggest the encoded protein may act as a transcriptional cofactor and be involved in the regulation of DNA transcription and replication. Alternatively spliced transcript variants have been described. [provided by RefSeq, Jul 2008]
Alternative Names PIR
Pirin
Molecular Weight(MW) 32, 36 kDa(Observed band size:36 kDa)
Cellular Localization Cytoplasm,Nucleus.
Application

WB

Western blot analysis of anti-PIR antibody. The sample well of each lane was loaded with 30 ug of sample under reducing conditions. After electrophoresis, proteins were transferred to a membrane. Then the membrane was incubated with rabbit anti-PIR antigen affinity purified polyclonal antibody and probed with a goat anti-rabbit IgG-HRP secondary antibody. The signal is developed using ECL Plus Western Blotting Substrate.

ICC/IF

ICC/IF analysis of PIR using anti-PIR antibody. PIR was detected in an immunocytochemical section of U2OS cells. Cy3-conjugated Anti-rabbit IgG Secondary Antibody (red) was used as secondary antibody. The tissue section was developed using Phalloidin-iFluor 488 Conjugated.

FC

Flow Cytometry analysis of HepG2 cells using anti-PIR antibody. Overlay histogram showing HepG2 cells stained with Rabbit polyclonal antibody to PIR (Blue line). To facilitate intracellular staining, cells were fixed with 4% paraformaldehyde and permeabilized with permeabilization buffer. The cells were blocked with 10% normal goat serum. And then incubated with rabbit anti-PIR Antibody (1:100). Fluoro488 conjugated goat anti-rabbit IgG (1:100) was used as secondary antibody. Isotype control antibody (Green line) was rabbit IgG (1:100) used under the same conditions. Unlabelled sample (Red line) was also used as a control.
