Coat the plate with 100ng immunizing antigen. The phospho-specific antibody,pbs only can specifically recognize phosphorylated peptide and enable quantitative detection over a wide dynamic range. Non-phosphorylated peptide shows no cross-reactivity, and the background signal remains low, indicating that the phospho-specific antibody,pbs only possesses high specificity and an excellent signal-to-noise ratio. Western blot with Anti-Phospho-NEDD4L(Ser448) antibody antibody on lysates of mouse lung tissue (1) .12% SDS-PAGE gel.Sample loading: 20μg /lane. Transfer the proteins onto a PVDF membrane (OM790003), and block it with TBST (OM750016) containing 5% BSA (OM297470) for one hour. Dilute the primary antibody with the antibody diluent (OM750012) at a ratio of 1:1000, and incubate it overnight at 4°C. Wash the membrane three times with TBST (OM750016), 5 minutes each time. At room temperature, dilute the secondary antibody, Goat Anti-Rabbit IgG(H&L)-HRP (OM643487), at a ratio of 1:10000 and incubate for one hour. HECT domain E3 ubiquitin ligases transfer ubiquitin from E2 ubiquitin-conjugating enzymes to protein substrates, thus targeting specific proteins for lysosomal degradation. The encoded protein mediates the ubiquitination of multiple target substrates and plays a critical role in epithelial sodium transport by regulating the cell surface expression of the epithelial sodium channel, ENaC.
