Product Profile
Product Name beta Catenin antibody [6F9]
Antibody Type Primary Antibodies
Product description Mouse monoclonal antibody [6F9] to beta CateninThe distinct peripheral cytosolic proteins, alpha, beta and gamma-catenin (102, 94 and 86 kD) found in many tissues bind to the conserved cytoplasmic tail domain of the cell-adhesion cadherins. Catenins link E-cadherin to other integral membrane or cytoplasmic proteins and are modulated by Wnt-1 protooncogene. They are good candidates for mediating transduction of cell-cell contact positional signals to the cell interior. Absence of alpha-catenin is found in certain tumor cell lines and reduced levels in certain human carcinomas. Beta-catenin binds directly to the cytoplasmic tail of E-cadherin. It binds to the N-terminus of alpha-catenin and interacts with the protein product of the tumor suppressor gene APC. This interaction involves a 15-aa repeat in the APC. Beta-catenin cell levels seem to be controlled by APC. The central core region of beta-catenin is involved in mediation of cadherin-catenin complex interaction with EGFR.
Immunogen Recombinant full length protein (Chicken).

Target beta Catenin
Tissue Specificity Recognises beta-catenin (94kD) in immunoblotting. Does not cross-react with plakoglobin.
Cellular Localization Cytoplasmic when it is unstabilized (high level of phosphorylation) or bound to CDH1. Translocates to the nucleus when it is stabilized (low level of phosphorylation).
Application

Application

Immunocytochemical staining of PFA-fixed MCF7 cells using anti-beta Catenin antibody [Red] and anti-TNIK antibody [Green]

Application

Cell line lysates were separated on SDS-PAGE and probed with 1:500 Monoclonal Anti-β-Catenin Clone: 6F9 (Cat. No. ). The antibody was developed using Goat Anti-Mouse IgG-Peroxidase and a chemiluminescent substrate. Lanes 1. HeLa 2. MBDK 3. MDCK 4. NRK 5. 3T3 6. Chicken fibroblasts

Application

MDCK cells were fixed and permeabilized with cold methanol followed by cold acetone. Fixed cells were stained with 1:500 Monoclonal Anti-β-Catenin Clone: 6F9 (Cat. No. ). The antibody was developed using Goat Anti-Mouse IgG, Atto-488 conjugate. Cells were counterstained with DAPI (blue) to stain nuclei.
Positive Control MDBK cells
Application Notes ICC: Use at an assay dependant dilution. IF: Use at a dilution of 1/1000. WB: Use at a dilution of 1/1000. Optimal dilutions/concentrations should be determined by the end user.
