Product Profile
Product Name Dynein Heavy chain antibody
Antibody Type Primary Antibodies
Product description Mouse monoclonal [440.4] to Dynein heavy chainEukaryotic cells rely on actin and microtubule-based protein ""motors"" to generate intracellular movements (3). These protein ""motors"" contain specialized domains that hydrolyze ATP to produce force and movement along a cytoskeletal polymer (actin in the case of myosin family and microtubules in the case of the kinesin family and dyneins). Dynein has been implicated in cytoplasmic motile functions, including chromosomal movement, retrograde organelle and axonal transport, the endocytic pathway, and the organization of the Golgi apparatus. In all cell types, dynein has the same basic structures and is composed of two or three distinct heavy chains (~450 kD), three intermediate chains (70-125 kD), and at least four light chains (15-25 kD) (4).
Immunogen Cytoplasmic full length native protein (purified from chick brain) (Chicken).

Target Dynein Heavy chain
Tissue Specificity The antibody identifies the cytoplasmic dynein heavy chain (440 kDa) in immunoblotting with dynein enriched preparations and in crude cell extracts of chicken brain and fibroblasts.
Cellular Localization Cytoplasmic. With intact mitotic chicken embryo fibroblasts, the labeling of the spindle with the antibody is clearly visible with diffuse cytoplasmic staining by immunofluorescence techniques (Wordeman L et al.).
Application

Application

Localization of cytoplasmic dynein to the mitotic spindle in dividing CEF cells. (C) Prometaphase. (F) Metaphase. (I) Anaphase. Immunofluorescent staining of cells using a mixture of Monoclonal Anti-Dynein (Heavy Chain), Clone No. 440.4 and Clone No. 440.1 (Cat. No.) after extraction with a detergent containing microtubule-stabilizing buffer.
Application Notes ELISA: Use at an assay dependant dilution. IF: Use at an assay dependant dilution. WB (colorimetric): Use at a dilution of 1/100 for chick brain extract. Optimal dilutions/concentrations should be determined by the end user.:
