Mouse monoclonal [2T15] to KaisoTranscriptional regulator with bimodal DNA-binding specificity. Binds to methylated CpG dinucleotides in the consensus sequence 5'-CGCG-3' and also binds to the non-methylated consensus sequence 5'-CTGCNA-3'. May recruit the N-CoR repressor complex to promote histone deacetylation and the formation of repressive chromatin structures in target gene promoters. Contributes to the repression of target genes of the Wnt signaling pathway. May also activate transcription of a subset of target genes by the recruitment of CTNND2.
Immunogen
Recombinant fusion protein (N-terminal), corresponding to amino acids 103 - 584 of mouse Kaiso.

Tested Applications
GSAIHCIPWB
For GSA: Use at an assay dependent dilution. Supershifts a specific methyl-CpG complex. For IHC: Use at a concentration of 1 μg/ml for positive nuclear staining in NIH3T3 cells fixed with 100% methanol. For IP: Use at an assay dependent dilution. For WB: Use at a concentration of 0.5 - 2 μg/ml on RIPA lysates from 3T3/A31 cells (ECL). Detects a band of approximately 97 kDa. Optimal dilutions/concentrations should be determined by the end user.:
Species Reactivity
HumanMouseDogRatHamster
Concentration
1mg/ml (Please refer to the vial label for the specific concentration)
3T3/A31 cell lysate. Add 2.5µl of 2-mercaptoethanol/100µl of lysate and boil for 5 minutes to reduce the preparation. Load 20µg of reduced lysate per lane for minigels.
