Product Profile
Product Name PPAR alpha antibody
Antibody Type Primary Antibodies
Product description Mouse monoclonal antibody [3B6/PPAR] to PPAR alphaPeroxisome proliferators include hypolipidemic drugs, herbicides, leukotriene antagonists, and plasticizers; this term arises because they induce an increase in the size and number of peroxisomes. Peroxisomes are subcellular organelles found in plants and animals that contain enzymes for respiration and for cholesterol and lipid metabolism. The action of peroxisome proliferators is thought to be mediated via specific receptors, called PPARs, which belong to the steroid hormone receptor superfamily. PPARs affect the expression of target genes involved in cell proliferation, cell differentiation and in immune and inflammation responses. Three closely related subtypes (alpha, beta/delta, and gamma) have been identified. This gene encodes the subtype PPAR-alpha, which is a nuclear transcription factor. Multiple alternatively spliced transcript variants have been described for this gene, although the full-length nature of only two has been determined. [provided by RefSeq, Jul 2008]

Application

Application

Flow cytometry analysis of PPAR alpha in Hela cells compared to an isotype control (blue). Cells were harvested, adjusted to a concentration of 1-5x10^6 cells/ml, fixed with 2% paraformaldehyde, washed with PBS, and incubated with PPAR alpha antibody [3B6/PPAR] at a dilution of 0.25 ug/test for 60 min at room temperature. Cells were then blocked in a solution of 2% BSA-PBS for 30 min at room temperature, incubated for 40 min at room temperature in the dark with a proper secondary antibody, and re-suspended in PBS for FACS analysis.

Application

Western blot analysis of PPAR alpha in 25 ug of HeLa, Jurkat and NIH-3T3 cell lysates. Proteins were transferred to a PVDF membrane and blocked with a blocking buffer at 4oC overnight. The membrane was probed with PPAR alpha antibody [3B6/PPAR] at a dilution of 1:1000 (HeLa and Jurkat) and 1:500 overnight at 4°C, washed in TBST, and probed with a proper secondary antibody for 1 hr at room temperature in the dark. Chemiluminescent detection was performed.
