Western blot analysis of BCAT1 using anti-BCAT1 antibody. The sample well of each lane was loaded with 30 ug of sample under reducing conditions. Lane 1: Raji whole cell lysates, Lane 2: MOLT-4 whole cell lysates, Lane 3: Hela whole cell lysates, Lane 4: rat pancreas tissue lysates, Lane 5: rat kidney tissue lysates, Lane 6: mouse kidney tissue lysates, Lane 7: mouse Neuro-2a whole cell lysates. After electrophoresis, proteins were transferred to a membrane. Then the membrane was incubated with rabbit anti-BCAT1 antigen affinity purified polyclonal antibody at a dilution of 1:1000 and probed with a goat anti-rabbit IgG-HRP secondary antibody. The signal is developed using ECL Plus Western Blotting Substrate.IHC

To facilitate intracellular staining, cells were fixed with 4% paraformaldehyde and permeabilized with permeabilization buffer. The cells were blocked with 10% normal goat serum. And then incubated with rabbit anti-BCAT1 Antibody at 1:100 dilution for 30 min at 20°C. Fluoro488 conjugated goat anti-rabbit IgG was used as secondary antibody at 1:100 dilution for 30 minutes at 20°C. Isotype control antibody (Green line) was rabbit IgG at 1:100 dilution used under the same conditions. | Gene Summary | This gene encodes the cytosolic form of the enzyme branched-chain amino acid transaminase. This enzyme catalyzes the reversible transamination of branched-chain alpha-keto acids to branched-chain L-amino acids essential for cell growth. Two different clinical disorders have been attributed to a defect of branched-chain amino acid transamination: hypervalinemia and hyperleucine-isoleucinemia. As there is also a gene encoding a mitochondrial form of this enzyme, mutations in either gene may contribute to these disorders. Alternatively spliced transcript variants have been described.
