Gene Full Name transgelin
Gene Summary This gene encodes a shape change and transformation sensitive actin-binding protein which belongs to the calponin family. It is ubiquitously expressed in vascular and visceral smooth muscle, and is an early marker of smooth muscle differentiation. The encoded protein is thought to be involved in calcium-independent smooth muscle contraction. It acts as a tumor suppressor, and the loss of its expression is an early event in cell transformation and the development of some tumors, coinciding with cellular plasticity. The encoded protein has a domain architecture consisting of an N-terminal calponin homology (CH) domain and a C-terminal calponin-like (CLIK) domain. Mice with a knockout of the orthologous gene are viable and fertile but their vascular smooth muscle cells exhibit alterations in the distribution of the actin filament and changes in cytoskeletal organization. Application

WB

Western blot analysis of transgelin/SM22/TAGLN using anti-transgelin/SM22/TAGLN antibody. The sample well of each lane was loaded with 30 ug of sample under reducing conditions. Lane 1: human HEPG2 whole cell lysates, Lane 2: rat stomach tissue lysates, Lane 3: mouse spleen tissue lysates. After electrophoresis, proteins were transferred to a membrane. Then the membrane was incubated with rabbit anti-transgelin/SM22/TAGLN antigen affinity purified polyclonal antibody at a dilution of 1:1000 and probed with a goat anti-rabbit IgG-HRP secondary antibody. The signal is developed using ECL Plus Western Blotting Substrate. A specific band was detected for transgelin/SM22/TAGLN at approximately 22 kDa. The expected band size for transgelin/SM22/TAGLN is at 22 kDa.

ICC/IF

ICC/IF analysis of transgelin/SM22/TAGLN using anti-transgelin/SM22/TAGLN antibody. transgelin/SM22/TAGLN was detected in an immunocytochemical section of Caco-2 cells. The section was incubated with rabbit anti-transgelin/SM22/TAGLN Antibody at a dilution of 1:100. Fluoro488 Conjugated Goat Anti-Rabbit IgG (Green) was used as secondary antibody. The section was counterstained with DAPI (Blue).

FC

Flow Cytometry analysis of HepG2 cells using anti-transgelin/SM22/TAGLN antibody. Overlay histogram showing HepG2 cells stained with Anti-transgelin/SM22/TAGLN Antibody (Blue line). To facilitate intracellular staining, cells were fixed with 4% paraformaldehyde and permeabilized with permeabilization buffer. The cells were blocked with 10% normal goat serum. And then incubated with rabbit anti-transgelin/SM22/TAGLN Antibody at 1:100 dilution for 30 min at 20°C. Fluoro488 conjugated goat anti-rabbit IgG was used as secondary antibody at 1:100 dilution for 30 minutes at 20°C. Isotype control antibody (Green line) was rabbit IgG at 1:100 dilution used under the same conditions. Unlabelled sample without incubation with primary antibody and secondary antibody (Red line) was used as a blank control.

IHC

IHC analysis of transgelin/SM22/TAGLN using anti-transgelin/SM22/TAGLN antibody. transgelin/SM22/TAGLN was detected in a paraffin-embedded section of human lung cancer tissue. Biotinylated goat anti-rabbit IgG was used as secondary antibody. The tissue section was incubated with rabbit anti-transgelin/SM22/TAGLN Antibody at a dilution of 1:200 and developed using Strepavidin-Biotin-Complex (SABC) with DAB as the chromogen.

IHC

IHC analysis of transgelin/SM22/TAGLN using anti-transgelin/SM22/TAGLN antibody. transgelin/SM22/TAGLN was detected in a paraffin-embedded section of human adrenal cortical adenocarcinoma tissue.

IF-P

IF analysis using anti- TAGLN antibody. detected in paraffin-embedded section of mouse heart tissue. The tissue section were stained using the Fluoro488-conjugated Anti-rabbit IgG Secondary Antibody (green) and counterstained with DAPI (blue).

IF-P

IF analysis using anti- TAGLN antibody. detected in paraffin-embedded section of human esophagus cancer tissue. The tissue section were stained using the Fluoro488-conjugated Anti-rabbit IgG Secondary Antibody (green) and counterstained with DAPI (blue).
