Western blot analysis of ME3 using anti-ME3 antibody. The sample well of each lane was loaded with 30 ug of sample under reducing conditions. Lane 1: monkey COS7 whole cell lysates, Lane 2: human HEL whole cell lysates, Lane 3: rat kidney tissue lysates, Lane 4: rat heart tissue lysates, Lane 5: rat brain tissue lysates, Lane 6: mouse kidney tissue lysates, Lane 8: mouse heart tissue lysates, Lane 9: mouse brain tissue lysates. After electrophoresis, proteins were transferred to a membrane. Then the membrane was incubated with rabbit anti-ME3 antigen affinity purified polyclonal antibody at a dilution of 1:1000 and probed with a goat anti-rabbit IgG-HRP secondary antibody. The signal is developed using ECL Plus Western Blotting Substrate. A specific band was detected for ME3 at approximately 67 kDa. The expected band size for ME3 is at 67 kDa.IHC
IHC analysis of ME3 using anti-ME3 antibody. ME3 was detected in a paraffin-embedded section of human placenta tissue. The tissue section was incubated with rabbit anti-ME3 Antibody at a dilution of 1:200 and developed using HRP Conjugated Rabbit IgG Super Vision Assay Kit with DAB as the chromogen.IF
IF analysis of ME3 using anti-ME3 antibody. ME3 was detected in a paraffin-embedded section of human placenta tissue. The tissue section was incubated with rabbit anti-ME3 Antibody at a dilution of 1:100. Fluoro594-conjugated Anti-rabbit IgG Secondary Antibody (red) was used as secondary antibody. Flow Cytometry analysis of HEL cells using anti-ME3 antibody. Overlay histogram showing HEL cells stained with (Blue line). To facilitate intracellular staining, cells were fixed with 4% paraformaldehyde and permeabilized with permeabilization buffer. The cells were blocked with 10% normal goat serum. And then incubated with rabbit anti-ME3 Antibody at 1:100 dilution for 30 min at 20°C. Fluoro488 conjugated goat anti-rabbit IgG was used as secondary antibody at 1:100 dilution for 30 minutes at 20°C. | Gene Summary | This gene encodes a nuclear protein that contains a p53 binding domain at the N-terminus and a RING finger domain at the C-terminus, and shows structural similarity to p53-binding protein MDM2. Both proteins bind the p53 tumor suppressor protein and inhibit its activity, and have been shown to be overexpressed in a variety of human cancers. However, unlike MDM2 which degrades p53, this protein inhibits p53 by binding its transcriptional activation domain. This protein also interacts with MDM2 protein via the RING finger domain, and inhibits the latter's degradation. So this protein can reverse MDM2-targeted degradation of p53, while maintaining suppression of p53 transactivation and apoptotic functions.
