Microtiter plate wells were coated with 100 ng/well coating antigen and incubated with serial dilutions of the antibody. Following HRP-conjugated secondary antibody and substrate development, OD630 was measured. The absorbance decreased progressively with increasing antibody dilution, yielding a sigmoidal titration curve that demonstrates the high antigen-binding activity of the polyclonal antibody.DB
Dot blot analysis was performed using Anti-Phospho-SKP2/p45 (Ser64) antibody to detect two peptide samples: lane 1 contains the phospho-peptide , and lane 2 contains the non-phospho-peptide. Transfer the proteins onto a NC membrane (OM790001), and block it with TBST (OM750016) containing 5% BSA(OM297470) for 1 hour. Dilute the primary antibody with the antibody diluent (OM750012) at a ratio of 1:1000, and incubate it at room temperature for 2 hours. Wash the membrane 3 times with TBST (OM750016), 5 minutes each time. At room temperature, dilute the secondary antibody, Goat Anti-Rabbit IgG(H&L)-HRP (OM643487), at a ratio of 1:10000 and incubate for 1 hour. Wash the membrane 3 times with TBST (OM750016) again, 5 minutes each time. Use ECL (OM625701) for luminescence. Exposure time: 30 s.WB

Transfer the proteins onto a PVDF membrane (OM790003), and block it with TBST (OM750016) containing 5% BSA (OM297470) for one hour. | Gene Summary | This gene encodes a member of the F-box protein family which is characterized by an approximately 40 amino acid motif, the F-box. The F-box proteins constitute one of the four subunits of ubiquitin protein ligase complex called SCFs (SKP1-cullin-F-box), which function in phosphorylation-dependent ubiquitination. The F-box proteins are divided into 3 classes: Fbws containing WD-40 domains, Fbls containing leucine-rich repeats, and Fbxs containing either different protein-protein interaction modules or no recognizable motifs. The protein encoded by this gene belongs to the Fbls class; in addition to an F-box, this protein contains 10 tandem leucine-rich repeats. This protein is an essential element of the cyclin A-CDK2 S-phase kinase. It specifically recognizes phosphorylated cyclin-dependent kinase inhibitor 1B (CDKN1B, also referred to as p27 or KIP1) predominantly in S phase and interacts with S-phase kinase-associated protein 1 (SKP1 or p19). In addition, this gene is established as a protooncogene causally involved in the pathogenesis of lymphomas.
