| Product description | Rabbit polyclonal to HDAC4Chromatin is a highly specialized structure composed of tightly compacted chromosomal DNA. Gene expression within the nucleus is controlled, in part, by a host of protein complexes which continuously pack and unpack the chromosomal DNA. One of the known mechanisms of this packing and unpacking process involves the acetylation and deacetylation of the histone proteins comprising the nucleosomal core. Acetylated histone proteins confer accessibility of the DNA template to the transcriptional machinery for expression. Histone deacetylases (HDACs) are chromatin remodeling factors that deacetylate histone proteins and thus, may act as transcriptional repressors. HDACs are classified by their sequence homology to the yeast HDACs and there are currently 2 classes. Class I proteins are related to Rpd3 and members of class II resemble Hda1p. |

| IF: Use at a dilution of 1/250, this is determined by indirect immunofluorescence using HEK 293T cells expressing recombinant mouse HDAC4. IP: Use at an assay dependent dilution. 10-20 μg of antibody immunoprecipitates HDAC4 from a RIPA extract of HeLa nuclei from 1 x 10^7 cells. WB: Use at an assay dependent dilution. Predicted molecular weight: 140 kDa. Additional weak bands may be detected using different extract preparations. Detection of HDAC4 is specifically inhibited with the immunizing peptide. A minimum working dilution of 1/1000 is determined by blotting using whole extracts of mouse NIH-3T3 cells. A minimum working dilution of 1/500 is determined by blotting using whole extracts of rat brain. Optimal dilutions/concentrations should be determined by the end user. | |
