NTRK1 (trkA), a Trk/Ror type protein kinase, is a high-affinity receptor for the nerve growth factor NGF. NTRK1 has been shown to regulate neural development and maintenance of neural network. Mutation in NTRK1 gene is responsible for congenital insensitivity to pain with anhidrosis (CIPA), an autosomal recessive hereditary disorder characterized by recurrent episodic fever, anhidrosis (inability to sweat), absence of reaction to noxious stimuli, self-mutilating behavior, and mental retardation. Rearrangements and fusion of NTRK1 gene to other genes have been implicated in colon and papillary thyroid cancers. Two protein isoforms have been identified.
