IHC image of MAGEA4 Monoclonal Antibody diluted at 1:200 and staining in paraffin-embedded human testis tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-mouse polymer IgG labeled by HRP and visualized using 0.05% DAB.IHC

Then 10% normal goat serum was Incubated to block non-specific protein-protein interactions followed by the antibody (1µg/1*106cells) for 45 min at 4°C. The secondary antibody used was FITC-conjugated Goat Anti-Mouse IgG(H+L) at 1/200 dilution for 35 min at 4°C. Isotype control antibody (green line) was mouse IgG1 (1µg/1*106cells) used under the same conditions. | Gene Summary | This gene is a member of the MAGEA gene family. The members of this family encode proteins with 50 to 80% sequence identity to each other. The promoters and first exons of the MAGEA genes show considerable variability, suggesting that the existence of this gene family enables the same function to be expressed under different transcriptional controls. The MAGEA genes are clustered at chromosomal location Xq28. They have been implicated in some hereditary disorders, such as dyskeratosis congenita.
