Application

Application

Western blot analysis of CX3CL1 in 0.25 μg of recombinant human CX3CL1 protein. Protein was transferred to a PVDF membrane and blocked with 5% Milk/TBST for at least 1 hour. Membrane was probed with CX3CL1 antibody [4C5] at a dilution of 1:1000 overnight at 4°C on a rocking platform. Membranes were washed in TBS-0.1% Tween-20 and probed with a HRP-conjugated secondary antibody. Membrane was washed and chemiluminescent detection was performed.

Application

ELISA analysis of human CX3CL1 was performed by coating a blank 96-well microtiter plate with 1μg/ml of recombinant CX3CL1 protein and incubating overnight. The plate was aspirated and blocked with block solution. The plate was washed then incubated for 2 hours with serial dilutions (10μg/ml) of CX3CL1 antibody [4C5]. The plate was washed and incubated with a proper secondary antibody, washed again, and 100μl per well of TMB substrate was added for 30 minutes at room temperature in the dark. The reaction was stopped with 0.16M sulfuric acid. Absorbances were read on a spectrophotometer at 450-550nm.
