CDK4, a CDC2/CDK-type protein kinase, regulates cell cycle G1 phase progression. CDK4 associates with and is regulated by D-type cyclins and the CDK inhibitor p16 (CDKN2A). Two alternatively spliced variants have been reported. CDK4 has been shown to phosphorylate the retinoblastoma protein, and mutations in the CDK4 gene have been linked to tumorigenesis in several types of cancers, including cutaneous malignant melanoma.
